3D culture의 Gold Standard ECM
Matrigel은 Engelbreth-Holm-Swarm(EHS) mouse sarcoma에서 extracellular matrix (ECM) 성분들을 분리한 제품으로 collagen V, Laminin 등의 ECM 성분을 포함하고 있습니다. 따라서 in vivo와 유사한 환경조성을 in vitro에서 구현할 수 있습니다. 또한, Matrigel은 in vivo tumor injection시 세포가 잘 안착할 수 있도록 도와주며, stem cell의 경우는 생체 내 환경과 유사한 ECM 성분을 제공하여 잘 성장하도록 도와줍니다.
Matrigel Matrix Handling Tips
How to Store
- 온도가 불안정한 환경에 보관하지 마세요. (ex. 자주 여닫는 냉동고, 냉동고 문, 서리가 자동으로 제거되는 냉동고)
- 최초 해동 후에는 냉동 호환 튜브에 aliquot 한 뒤, -70°C 또는 -20°C에서 보관하세요.
How to Thaw
- Matrigel 매트릭스는 10°C 이상에서 gel화가 시작됩니다.
- 2°C에서 8°C에서 하룻밤 동안 해동하세요. 해동하는 동안 바이알 주변에 얼음이 있어야 합니다.
How to Dilute
- 해동된 Matrigel 매트릭스를 얼음이 담긴 serum free media 혹은 PBS에 희석하세요.
- 얼음 위에서 부드럽게 흔들거나 피펫으로 위아래로 펌핑하여 고르게 분포되도록 합니다.
- 항상 Matrigel 매트릭스는 표준 부피 희석이 아닌 단백질 농도를 기준으로 희석하세요.
How to Coat
- 세포 부착 및 증식 응용을 위한 경우: 얇은 Matrigel 매트릭스 층을 사용하세요. 표면을 최소 50μㅣ/cm2 Matrigel 매트릭스로 코팅하세요.
- 3D 세포 배양 응용을 위한 경우: 두꺼운 Matrigel 매트릭스 층을 사용하세요. 최소 150~200 μL/cm² Matrigel 매트릭스를 사용하거나, 최소 7 mg/mL 농도의 Matrigel 매트릭스를 50 μL 사용하여 3D dome을 형성하세요.
- 배지를 코팅하는 동안에는 얼음 위의 Corning CoolRack™ 또는 CoolBox™의 얼음에 보관하세요.
- 3D dome을 형성할 때는 TC 처리된 배양 용기를 미리 인큐베이터에 넣기 전에 따뜻하게 유지하여 dome이 폴리머화 되도록 도와주세요.
